Water Activity as a Primary Indicator of Moisture in Lyophilized Products

Is Karl Fischer titration telling you the whole moisture story?

Karl Fischer titration reports total water content, but it cannot separate the “active” water that drives hydrolysis and microbial growth from water bound tightly within the product matrix. It is also destructive, which limits how many vials can realistically be tested and leaves little visibility into vial-to-vial or batch-to-batch variability.

This article shows how non-destructive FMS-Water Activity headspace analysis, paired with a moisture sorption isotherm generated on a Dynamic Vapor Sorption (DVS) instrument, can report both water activity and total moisture content from a single measurement, consistent with USP <922>. Testing across 87 vials from four production batches of a lyophilized protein product shows the approach reliably predicts Karl Fischer results, including for vials deliberately spiked with additional moisture.

For formulation scientists, analytical development groups, and quality teams accountable for moisture as a critical quality attribute, this is a practical look at replacing single-point destructive testing with a non-destructive method suited to both release and stability programs.
